This book Deals with Determination of carbohydrate antigen 15-3 (CA15-3) and carbohydrate antigen 19-9 (CA 19-9) levels in sera of patients with benign and malignant breast tumors , Development of IRMA assay for the determination of CA15-3 and CA19-9 from cytosolic tissues of benign and malignant breast tumors ,Characterization of the binding of 125I-anti CA15-3 antibody with isolated human - CA15-3 in benign and malignant breast tumors, such as those of binding capacity and the effect of various factors (pH, temperature, time, halides, salts, CA15-3 and its antibody concentration) ,Determination of kinetic and thermodynamic parameters of the binding of partially purified CA15-3 with its specific antibody and Spectroscopic studies on (125I-anti CA15-3 antibody / CA15-3) complex in breast tissue . Further more the level of sera CA 15-3 in patients with benign and malignant breast tumors (preoperative) was measured by Immunoradiometric Assay (IRMA). Data analysis showed, concentrations of CA 15-3 were significantly higher in pre-and post-menopausal malignant breast tumors (P< 0.0001) and significantly lower in benign breast tumors, compared with healthy subjects.The results obtained revealed higher incidence of CA 15-3 in two groups of malignant breast tumors than those in benign breast tissues and in the supernatant fraction more than the pellet fraction .The binding of 125I -anti CA 15-3 antibodies with CA 15-3 was studied in three groups: benign breast tumor (Fibroadenoma), pre-and post- menopausal malignant breast tumors (IDC). The optimum conditions observed for the binding were as follows:CA 15-3 in concentration tissue homogenate 100?g. mL-1 for groups II and I while it was 200 ?g.mL-1 for group III. 125I -anti CA 15-3 antibody concentrations: 0.175?g.mL-1 for group I and II, whereas it was 0.140 ?g.mL-1 for group III. Temperatures of incubation were: 45oC for groups I and III, 15 oC for group II, while time of incubation was 90 min for both group I and III, 30 min. for group II. The optimum pH was 7.0 for group I, 7.6 for group II and 7.8 for group III. The use of different halides was shown to increase the binding between CA 15-3 and 125I -anti CA 15-3 antibody in both group II and III, while inhibition occurred on the binding in group I.
